【 摘 要 】
PresentstudydemonstratestheimmobilizationofacetonefractionatedIpomoeabatata(sweetpotato)βamylaseonaninorganicsupport,Celite-545bysimpleadsorptionmechanism.Theadsorbedenzymeexhibitedanactivityyieldof244Ug-1ofthematrixwitheffectivenessfactor‘η’0.83.InteractionbetweenCelite-545andenzymewasconfirmedbyfouriertransforminfraredspectroscopyandatomicforcemicroscopy.Thebindingefficiencyofenzymetothesupportwasanalyzedbyelutingitwith1.0MNaCl.BothsolubleandimmobilizedβamylaseexhibitedsamepHoptimawhiletemperatureoptimumofimmobilizedenzymewasshiftedfrom50oCto60oC.Theimmobilizedβamylasepreparationwassuperiortothefreeenzymeinhydrolyzingstarchinabatchprocess:ithydrolyzedstarchto88%and96%at40oCand50oC,respectivelywhereassolubleenzymehydrolyzedonly83%and80%ofstarchundersimilarexperimentalconditions.Theimmobilizedβamylaseretained84%ofitsoriginalactivityafter30daysstorageat4oC,whilethesolubleenzymeshowedonly41%oftheinitialactivityunderidenticalconditions.Immobilizedβamylaseretained79%activityevenafterits7threpeateduse.
