【 摘 要 】

Themostefficientwayofproducingandsupplyingproperrawmaterialdesiredbythepastaindustryisthedevelopmentofhighqualitydurumwheatvarieties.Therefore,qualityofTurkishdurumwheatvarietiesshouldbeimprovedusingmodernbreedingmethodswithoutadverselyaffectingtheiryields.Inthisstudy,importantgenes(y-gliadin45andLMW-2glutenin)affectingthequalityofpastaproductsweretransferredtoaTurkishdurumwheatvariety,Selçuklu-97,inabackcrossbreedingmethodincombinationwithmarkerassistedselection(MAS).ACanadiandurumwheatcultivarwithhighquality,Kyle,wasusedasthedonorparent.EachofF1andbackcross(BC)plantswasbackcrossedfourtimestotherecurrentparentandinallofthegenerations,backcrossedplantscarryingthetargetedQTLswereselectedbytheMAS.TheMASmethodwasemployedincombinationwithembryocultureandrapidplantgrowthinacontrolledgreenhouseconditions.Inidentifyingandtransferringprocessesofthegeneregions,molecularDNAmarkers(SSR,STSandGAG)wereemployedwithA-PAGEandSDS-PAGEmethods.A-PAGEwasusedforselectionofy-gliadin45,SDS-PAGEforselectionofLMW-2glutenins,andfourSSRprimers(Xgwm550,Xgwm608,Stm553actc,Stm542acag),oneSTSprimerandonePCRprimer(GAG5-6)linkedtoGli-B1andGlu-B3lociforselectionofy-gliadin45andLMW-2gluteninseitheralltogetheroralternatively.Asaresult,thestudywascompletedinthreeyearsinsteadofsixyearsrequiredinaclassicalbackcrossbreedingstudy,meaningabout50%timesaving,andobtainedahighqualitycandidatevariety.
