【 摘 要 】
Thepresentstudyhasbeenconductedon120bacterialisolatescollectedfromspoiledstarchymaterialslikewheatflourandbreaddough,soilandspoiledsourmaterialsliketomatoes,citrusfruitsandpearsetc.Theseisolateswerescreenedfortheproductionofextracellularacidophilicandthermostableamylases.Fiveisolatesexhibitedgoodamylaseactivity.PreliminaryidentificationoftheseisolatesshowedthattheyallbelongtothegenusBacillus,sotheyweredesignatedasBacillussp.JAR-4,JAR-26,JAR-29,JAR-53andJAR-76.IsolateJAR-26showingmaximumstarchhydrolysiswasfurthersubjectedtobiochemicaltestingandonthebasisofresultsitwasdesignatedasBacilluslicheniformisJAR-26.Itwasabletogrowupto550CatpH5.5andshowedremarkableliquefactionaswellassaccharificationactivity.TheoptimumperiodformaximumcellgrowthandamylaseproductionincaseofBacillussp.JAR-26wasfoundtobe36hat450C.Theoptimumtemperatureforenzymeassaywas850CatpH5.5.Theenzymewasalsothermostableas100%activitywasobservedat850Cand55%activityretainedat1000Cand48%activityat1040Cwhenheatedfor30min.
