【 摘 要 】
Inthisstudy,fababean(Viciafaba)seedlingswereusedasbioindicatortodeterminegenotoxiceffectofsyntheticdyescurrentlyusedasfoodcoloradditivesinmanycountries.Novelshort-termassaysarerequiredtosubstantiatethebatteryofassessmentmethodsforevaluatingthegenotoxicityofcandidatesubstances.Therefore,anattempthasbeenmadetoevaluaterandomlyamplifiedpolymorphicDNA(RAPD)analysisforitspotentialtoestablishgenotoxiceffectofcoloredfood.Forthepreliminaryassessment,thisstudycomparedtheeffectsoccurringatmolecularlevelsinViciafabaexposedtocoloredfoodatconcentrationsintherangeof0.2%to18.2%.ThequalitativemodificationsarisinginrandomamplifiedpolymorphicDNA(RAPD)profilesasameasureofDNAeffectswerecomparedwithcontrol.ResultssuggestedthattreatmentsoftheabovetestfoodsamplesreflectingchangesinRAPDprofiles.ChangesinRAPDpatternsincludedvariationinbandintensity;lossofnormalbandsandappearanceofnewbandscomparedwithcontrol.TheseresultsindicatedthatDNApolymorphismsdetectedbyRAPDanalysiscouldbeusedasaninvestigationtoolforenvironmentaltoxicologyandasausefulbiomarkerassayforthedetectionofgenotoxiceffectsoffooddyes.Inconclusion,themeasurementofparametersatmolecularlevelsisvaluableforinvestigatingthespecificeffectsofagentsinteractingwithDNA.Ultimately,thismethodologymayallowtheecotoxicologicalexaminationofthelinkbetweenmolecularalterationsandmeasurableadverseeffectsathigherlevelsofbiologicalorganization.
