【 摘 要 】
Melatoninhasbeenshowntobeaneffectiveantioxidantinanumberofexperimentalmodelsbothinvitroandinvivo.However,limitedinformationisavailableonmonitoringtheeffectofmelatoninontheantioxidantenzymestatusintheliverinsinglecircadianrhythm.Tomonitortheeffectofmelatoninonantioxidantenzymeactivity,weusedSwissalbinoinbredmalemiceasamodelsystem.Thecrudeenzymefractionswerepreparedfromfreshlyisolatedliversat0,2,and24hoursaslightcycleand4,8,and12hoursasdarkcyclespecimens,postinjectionofmelatonin.TheinjectionofmelatoninresultedinastatisticallysignificantdecreaseinactivityofSODcomparedtocontrolgroup(p<0.01).However,harvestingtimedidnotaffectSODlevelssignificantly.TheactivityofCATwassignificantlydecreasedbymelatoninandthisdecreasewasclearlyrelatedtothetimeofharvest(p<0.01).InhibitiononSODactivitystartedatthebeginningofthedarkcycleofthecircadianrhythm,whileCATactivitystartedtodecreaseatthatpoint.TheinhibitionofCATreacheditsmaximumat8hourpostinjection.After12hourpostinjection,thebeginningofthelightcycleperiod;CATactivityturnedbacktothecontrollevel.Weconcludedthatbasedoncontroldata,internalmelatoninreleaseincreasesSODandCATactivity.Theexternalmelatonintreatmentdirectlyinhibitsmostly•OHradicalsandH2O2aswellassingletoxygen(¹O²).
