【 摘 要 】

Duringconstructionofgenomiclibrariesfromvariousbacteria,weexperiencedconsiderablydifferenttransformationefficienciesamongthestrainsofelectrocompetentEscherichiacolicellsandinvestigatedtheeffectoftypestrainonefficiencyoftransformation.Amongthefivestrainstested,EscherichiacoliDH10BandTOP10werefoundtobethemostsuitablestrainsforconstructionofgenomiclibrariesinourlaboratory.ThisstudyhighlightedthefactthatalthoughthesecommerciallyavailableEscherichiacolistrainsarewellcharacterized,theytendtodifferentiateandloosetheirtransformationabilities.Thismaybeduetothefactthatthenumberofpassagestheyhavegonethroughaffecttheirmembranestructuresandpermeability.Therefore,everylaboratoryshouldtesttheirstrainsforcompetencybeforetheyusethemintheircloningstudies.
