Effects of Chronic Exposure to Cobalt Chloride on the Fertility and Testes in Mice

【 摘 要 】

Theeffectoftwelveweeksingestionofcobaltchloride(CoCl2)wasinvestigatedonthefertilityofadultmaleSwissmice.Sexuallymaturemalemicewereexposedto200,400and800ppmcobaltchloridedissolvedindrinkingwater.Basedonfluidconsumption,exposedanimalsreceivedanaverageof25.66±2.34,46.91±4.78and93.01±6.76mg/kg/daycobaltchloride,respectively.Aftertheexposureperiod,thesemaleswerematedwithuntreatedfemalemice.Fertilitywassignificantlyreducedinmalesthatingestedcobaltchloride.ThenumberofpregnantfemalesandthenumberofimplantationsitesweresignificantlyreducedinfemalesmatedwithmalesthathadingestedCoCl2ataconcentrationof400or800ppm.ThetotalnumberofresorptionsandthenumberoffemaleswithresorptionsweresignificantlyincreasedinfemalesmatedwithmalesthathadingestedCoCl2atallconcentrations.Ontheotherhand,thenumberofviablefetuseswasdecreasedinfemalesimpregnatedbyexposedmalesatthethreeconcentrations.Absoluteepididimalweight,andtherelativeorabsolutetestesweightsweresignificantlyreducedinmalesthatingestedcobaltchlorideataconcentrationsof400or800ppm.Incontrast,absoluteandrelativeweightsoftheseminalvesiclesweresignificantlyincreased.Furthermor,epididymalandspermcountsanddailyspermproductionweresignificantlydecreasedinmalesthatingestedCoCl2.HistologicalevaluationofthetestesrevealedseveralabnormalitiesincludinghypertrophyoftheinterstitialLeydigcells,congestedbloodvessels,degenerationofthespermatogonialcellsandnecrosisofboththeseminiferoustubulesandtheinterstitialtissue.
