ThepresentreviewdescribesTILLING(TargettingInducedLocalLesionsInGenomes)whichisageneralreversegeneticstrategythatworkswithamismatch-specificendonucleasetodetectinducedornaturalDNApolymorphismsingenesofinterest.Withhigh-throughputTILLING,rapidandlow-costdiscoveryofinducedpointmutationsinpopulationsofchemicallymutagenizedindividualsfromdiverseorganismsbecomespossible.Otheradvantagesareitsindependenceofgenomesize,reproductivesystemorgenerationtime.TILLINGyieldsatraditionalallelicseriesofpointmutationswhichmakeitvaluableforessentialgenes,wheresublethalallelesarerequiredforphenotypicanalysis.CurrentadvantagesofTILLINGhavemadeitanappropriatechoiceforthedetectionofbothinducedandnaturalvariationinseveralplantspecies.Here,wereviewrecentprogressinthistechnologyfortheresearchersofplantmutationanalysisandgenomicsera.
