Sub-Cellular Damage of Hepatocytes Caused by Different Doses of Diclofenac Sodium in Rabbit

【 摘 要 】

Diclofenacsodiumisapotentanalgesicandanti-inflammatorydrugthatisextensivelyprescribedintreatmentofrheumatoidarthritis,postoperativepain,andchronicpainassociatedwithcancer.Thepresentstudywasdesignedtoelucidatethequalitativeandquantitativechangesinroughendoplasmicreticulumofhepatocytesafterrecommendedsingle,doubleandtripletherapeuticdoseofDiclofenacsodiuminrabbits.Theaimofstudyistominimizetheindiscriminateuseofthisdrugincommunityandamongphysicians.ExperimentalstudywascarriedatDowUniversityofHealthSciences(DUHS)andSindhInstituteofUrologyandTransplantation(SIUT)fromMarch2009toJune2010.Eightyeighthealthyanimalsofthreemonthsageand900~1000gmbodyweightwereisolatedfromtheanimalhouseofDUHS.TheseanimalsweredividedintofourgroupscategorizedintoGroupA,GroupB,GroupC,GroupD,eachcontainingtwentytwoanimals.Diclofenacsodiumwereadministeredintraperitoneallywiththedailydosesof2,4,and6mg/kgbodyweightfor14consecutivedaysinGroupsB,CandDwhilethecontrolgroup(GroupA)receivednormalsaline.Animalsweresacrificedondayfifteenandliverswereremovedandfixedin4%gluterldehyde.Theywereprocessedforelectronmicroscopyandexaminedundertransmissionelectronmicroscope.Datawascollectedandsubjectedforstatisticalanalysisa‘P’valuelessthan0.05wastakenassignificant.ItwasobservedthatDiclofenacsodiumproducessignificantchangesinhepatocytes.Therewasde-granulationandswellingofthecisternaeofRoughEndoplasmicReticulum(RER)whenthedoseisdoubledandthesechangeswerehighlysignificantwhenthedoseisincreasedtothreetimesthetherapeutic.
