【 摘 要 】
TelomeresaretheendsofthechromosomesandrepresentrepeatedDNAsequencesandproteincomplexes.Telomeresshortenwitheachcelldivision,whichlimitstheproliferationpotentialofcells.Currentlythereisintensivedevelopmentintheclinicalapplicationoftelomerelength.Additionalcharacteristicsofstemcellproliferationactivitybytelomerelengthcanbeaninformativeindicatoroftransplantquality.Weanalyzed15cordbloodsamplesusingflow-FISH,ELISA,hematologicalanalyzer,andnumberofCD34+CD45dimcells.TheaveragetelomerelengthofCBcells’leukocytesfractionwas20.1±4.8%respectiveto1301controlcells(T-celllymphoblasticleukemia).LinearcorrelationanalysisdidnotfindanyconnectionbetweenthetelomerelengthoftheCBcells’leukocytefraction,andCD34+CD45dimcellnumbers(r=-0.49),whichisinlinewithpublisheddata.However,theaveragetelomerelengthoftheCBleukocytefractionpositivelycorrelateswithmonocyteconcentration(r=0.65).Therewerecomparativelylongtelomeres(23.6and23.9%)intwosamplesoftheCBcell’leukocytefractionwithoutanycontamination.Thisdataconfirmsthattheflow-FISHmethodissuitablefortheestimationoftelomerelengthoftheleukocytefractionofCBcells.AveragetelomerelengthoftheleukocytefractionofCBcellsobliquelyshowsCBsamplequality,duetoapositivecorrelationwithmonocyteconcentrationandcontaminationabsence.
