【 摘 要 】

Umbilicalcordbloodisasourceofhematopoieticstemcellsfortransplantationintreatmentofmanydevastatingblooddiseasesandgeneticmetabolicdisorders.Transplantationsuccessdependsonhematopoieticstemandprogenitor(calledcolony-formingunits(CFU))cellnumbers.Aim:Toanalyzethecolony-formingactivityofumbilicalcordbloodsamplesbeforeandafterfreezing(cryopreservation).Methods:TheleukocytefractionofcordbloodwasderivedusingRubinstein’sdoublecentrifugationmethod.Thecolony-formingactivityofcordbloodwasdetermined.ThemononuclearcellfractioncontaininghematopoieticprogenitorswasincubatedinthemethylcellulosemediumMethoCultH4435(StemCellTechnologies)over14days.NumbersofCFU-E,BFU-E,CFU-GM,andCFU-GEMMwereestimatedusingstandardmethods.Results:124samplesofcordbloodweretested.CFU-Emadeup6.2%ofallgrowncolonies,BFU-E55.8%,CFU-GM33.1%,andCFU-GEMM4.5%.AmoderatepositivecorrelationbetweenlivingCD34+stemcellnumbersandthetotalcolonynumberwasfound(r=0.676;plt;0.01).ThemostsignificantpositivecorrelationbetweentheCD34+cellnumberandcolonytypewasfoundinthecaseofBFU-E(r=0.633;plt;0.01),incomparisonwiththeothercolonytypes:CFU-E,CFU-GM,andCFU-GEMM(r=0.418;r=0.452,andr=0.42;plt;0.01,respectively).TherewasaweaktendencytowardcolonynumberdecreaseafterthawingascomparedtotheCFUnumbersbeforefreezinginthesamesamples,howeverasignificantdifferencewasnotfound.
