【 摘 要 】

Heparanaseisanendoglucuronidasethatcleavesheparansulfatechains.Heparanaseexpressionisupregulatedinmanytypesofcancer,particularlyinthosetumorsexhibitinganaggressivephenotype.Inmyelomapatients,heparanaseisoftenpresentinhighlevelsinthebonemarrowwhereitisassociatedwithenhancedangiogenesisandpoorpatientprognosis.Usinginvitroandinvivomodelsofmyeloma,wehaveprobedthemechanismofheparanaseactioninmyelomaanddiscoveredthattheenzymeactivelypromotestumorgrowth,angiogenesis,metastasis,andosteolysis.Thisoccursthroughheparanase-mediatedupregulationofexpressionofanumberofproteinsknowntodriveanaggressivetumorphenotypeincludingHGF,MMP-9,VEGF,andRANKL.Inaddition,heparanaseenhancessheddingofsyndecan-1(CD138),aheparansulfateproteoglycanpresentathighlevelsonthemyelomacellsurface.Thisisimportantbecausetheheparansulfatechainspresentontheshedsyndecan-1bindtoeffectorproteinssuchasHGFandVEGFandenhancetheirabilitytopromotesignaling.Thisfurtherstimulatesmyelomagrowthandangiogenesis.Whenmicebearingmyelomatumorsweretreatedwithinhibitorsofheparanaseenzymaticactivity,tumorgrowthandprogressionweresignificantlyattenuated.Togethertheseresultsdemonstratethatheparanaseplaysacriticalroleinenhancingmyelomaprogressionandthattargetingheparanasemaybeaviabletherapeuticstrategyfortreatingmyelomaandothercancers.
