【 摘 要 】

Background:Porphyromonasgingivalisinhibitsoralepithelialwoundhealinginvitromorestronglythanotheroralbacteria,butitisunknownwhyP.gingivalisissuchapotentinhibitorofwoundhealing.Objective:Therefore,theaimofthisstudywastoinvestigatetheinfluenceofmajorvirulencefactorsofP.gingivalisonwoundhealinginaninvitrowound-healingmodel.Theinfluenceofthecapsularpolysaccharide,theArg-andLys-gingipains,themajorfimbriaeandlipopolysaccharide(LPS)wasinvestigated.Design:AstandardizedscratchwasmadeinaconfluentlayerofhumanoralepithelialcellsHO-1-N-1.TheepithelialcellswerethenchallengedwithdifferentconcentrationsofseveralP.gingivaliswild-typestrainsandknockoutmutants.Closureofthescratchwasdeterminedafter17handcomparedtocontrolconditionswithoutbacteria.Results:TheP.gingivalisstrainsATCC33277,W83,andW50significantlyinhibitedwoundhealing.Thepresenceofacapsularpolysaccharideloweredsignificantlytheinhibitionofepithelialcellmigration,whilegingipainactivitysignificantlyincreasedtheinhibitionofcellmigration.LPSandthemajorfimbriaedidnotinfluenceepithelialcellmigration.NoneofthetestedP.gingivalisstrainscompletelypreventedtheinhibitionofcellmigration,suggestingthatothercharacteristicsofP.gingivalisalsoplayaroleintheinhibitionofwoundhealing,andthatfurtherresearchisneeded.Conclusions:ThecapsularpolysaccharideandtheArg-andLys-gingipainsofP.gingivalisinfluencedthecapacityofP.gingivalistohinderwoundhealing,whileLPSandthemajorfimbriaehadnoeffect.Keywords:gingipains;capsularpolysaccharide;P.gingivalis;LPS;cellmigrationessay;oralepithelialcells;re-epithelialization(Published:2July2015)Citation:JournalofOralMicrobiology2015,7:27543-http://dx.doi.org/10.3402/jom.v7.27543
