【 摘 要 】
Background:Biofilmsareorganizedcommunitiesofmicroorganismsembeddedinaself-producedextracellularpolymericmatrix(EPM),oftenwithgreatphylogeneticvariety.Bacteriainthesubgingivalbiofilmarekeyfactorsthatcauseperiodontaldiseases;amongthesearetheGram-negativebacteriaFusobacteriumnucleatumandPorphyromonasgingivalis.TheobjectivesofthisstudyweretocharacterizethemajorcomponentsoftheEPMandtotesttheeffectofdeoxyribonucleaseI(DNaseI)andproteinaseK.Methods:F.nucleatumandP.gingivalisbacterialcellsweregrownindynamicandstaticbiofilmmodels.TheeffectsofDNaseIandproteinaseKenzymesonthemajorcomponentsoftheEPMweretestedduringbiofilmformationandonmaturebiofilm.Confocallaserscanningmicroscopywasusedinobservingbiofilmstructure.Results:Proteinsandcarbohydrateswerethemajorcomponentsofthebiofilmmatrix,andextracellularDNA(eDNA)wasalsopresent.DNaseIandproteinaseKenzymeshadlittleeffectonbiofilmsintheconditionsused.Intheflowcell,F.nucleatumwasabletogrowinpartiallyoxygenatedconditionswhileP.gingivalisfailedtoformbiofilmaloneinsimilarconditions.F.nucleatumsupportedthegrowthofP.gingivaliswhentheyweregrowntogetherasdualspeciesbiofilm.Conclusion:DNaseIandproteinaseKhadlittleeffectonthebiofilmmatrixintheconditionsused.F.nucleatumformedbiofilmeasilyandsupportedthegrowthofP.gingivalis,whichpreferredanaerobicconditions.Keywords:Subgingivalbiofilm;extracellularpolymericmatrix;Fusobacteriumnucleatum;Porphyromonasgingivalis;staticanddynamicbiofilmmodels;confocallaserscanningmicroscopy(Published:29Janaury2013)Citation:JournalofOralMicrobiology2013,5:20015-http://dx.doi.org/10.3402/jom.v5i0.20015
