Differential transcription of virulence genes inAggregatibacter actinomycetemcomitansserotypes

【 摘 要 】

Background:Aggregatibacteractinomycetemcomitansserotypesareclearlyassociatedwithperiodontitisorhealth,whichsuggestsdistinctstrategiesforsurvivalwithinthehost.Objective:Weinvestigatedthetranscriptionprofileofvirulence-associatedgenesinA.actinomycetemcomitansserotypeb(JP2andSUNY465)strainsassociatedwithdiseaseandserotypea(ATCC29523)strainassociatedwithhealth.Design:Bacteriawereco-culturedwithimmortalizedgingivalepithelialcells(OBA-9).Theadhesionefficiencyafter2hoursandtherelativetranscriptionof13geneswereevaluatedafter2and24hoursofinteraction.Results:AllstrainswereabletoadheretoOBA-9,andthiscontactinducedtranscriptionofpgAforpolysaccharidebiosynthesisinalltestedstrains.GenesencodingvirulencefactorsasOmp29,Omp100,leukotoxin,andCagE(apoptoticprotein)weremoretranscribedbyserotypebstrainsthanbyserotypea.ltxAandomp29,encodingtheleukotoxinandthehighlyantigenicOmp29,wereinducedinserotypebbyinteractionwithepithelialcells.Factorsrelatedtocolonization(aae,flp,apaH,andpgA)andcdtBwereupregulatedinserotypeastrainafterprolongedinteractionwithOBA-9.Conclusion:Genesrelevantforsurfacecolonizationandinteractionwiththeimmunesystemareregulateddifferentlyamongthestrains,whichmayhelpexplainingtheirdifferencesinassociationwithdisease.Keywords:Aggregatibacteractinomycetemcomitans;geneexpression;epithelialcell;infection(Published:22October2013)Citation:JournalofOralMicrobiology2013,5:21473-http://dx.doi.org/10.3402/jom.v5i0.21473
