【 摘 要 】
Background:Directmicroscopy,anaerobiccultureandDNA-DNAhybridizationhavepreviouslydemonstratedanassociationbetweenmicroorganismsandosteoradionecrosis(ORN).Thepurposeofourstudywastousecultureindependentmoleculartechniquestodetectbacteriainnecroticbonelesionsofthemandibleafterradiationtherapy.Design:BacterialDNAwasextractedfromeightdeepmedullarspecimensfromresectedmandibles(sixcases),includingonepatientwithrelapse.16SrRNAgeneswerePCRamplified,cloned,transformedintoEscherichiacoliandsequencedtodeterminespeciesidentityandclosestrelatives.Results:Fromtheanalysisof438clones,59predominantspeciesweredetected,27%ofwhichhavenotbeencultivated.ThepredominantspeciesdetectedfromradionecroticmandibleswereCampylobactergracilis,Streptococcusintermedius,Peptostreptococcussp.oralcloneFG014,unculturedbacteriumcloneRL178,Fusobacteriumnucleatum,andPrevotellaspp.ThestudydemonstratedintersubjectvariabilityofthebacteriapresentinORN.Incontrasttothediversebacterialprofiledetectedinprimaryinfection,onlyafewmembersoftheoralindigenousflorawereidentifiedfromtherelapsecase.Conclusions:DiversebacterialprofilesinspecimensofORNinmarrowspacesofthemandibleweredetectedbycultureindependentmoleculartechniques.Tobetterunderstandthepathogenesisandtoimprovethetherapyoftheinfection,detectionofallmembersofthecomplexbacterialfloraassociatedwithORNisnecessary.Keywords:osteoradionecrosis;bacteria;16SrRNAgenes(Published:13July2010)Citation:JournalofOralMicrobiology2010.2:5072-DOI:10.3402/jom.v2i0.5072*TheChiefEditorIngarOlsen,hasnothadanypartinthereviewanddecisionprocessforthispaper.
