【 摘 要 】
Nitricoxide(NO)issuggestedtoplayanimportantroleinprimaryheadaches.IthasbeenproposedthatreleaseofNOfromsatelliteglialcells(SGCs)ofthetrigeminalganglion(TG)couldcontributetothepathogenesisoftheseheadaches.TheprincipalaimofthisstudywastoinvestigateifthephosphodiesteraseinhibitorIbudilast(Ibu)and1#945;,25-dihydroxyvitaminD3(VitD3)couldinterferewithNOreleasefromtrigeminalSGCs.SinceglutamateisreleasedfromactivatedTGneurons,theabilityofglutamatetoalterNOreleasefromSGCswasalsoinvestigated.Tostudythis,weisolatedSGCsfromtheTGofadultmaleSprague-Dawleyrats,provokedNOreleasefromSGCswithforskolin(FSK;0.1,1,10#181;M),andexaminedtheeffectofgradedconcentrationsofIbu(1,10,100#181;M),VitD3(5,50,500nM),andglutamate(10,100,1000#181;M).OurresultsindicatethatbothIbuandVitD3arecapableofattenuatingtheFSK-mediatedincreasedNOreleasefromSGCsafter48hoursofincubation.Lowerglutamateconcentrations(10and100#181;M)significantlydecreasedNOreleasenotonlyunderbasalconditionsafter24and48hours,butalsoafterSGCswerestimulatedwithFSKfor48hours.Inconclusion,NOreleasefromSGCsharvestedfromtheTGcanbeattenuatedbyglialmodulatorsandglutamate.AsNOisthoughttoincreaseTGneuronexcitability,thefindingssuggestthattargetingSGCsmayprovideanoveltherapeuticapproachformanagementofcraniofacialpainconditionssuchasmigraineinthefuture.(IJPPP1310001).
