【 摘 要 】
FragileXsyndrome(FXS)iscausedbythelossoffragileXmentalretardationprotein(FMRP).ThedeficiencyofGABAAreceptors(GABAARs)isimplicatedinFXS.However,theunderlyingmechanismsremainunclear.ToinvestigatetheeffectofFMRPonGABAARs,wetransfectedFMRPcDNAsinratcorticalneurons.WemeasuredtheproteinexpressionofGABAARsandphosphatasePTEN,andrecordedGABAAR-mediatedwhole-cellcurrentsinthetransfectedneurons.WeshowthatthetransfectionofFMRPcDNAscausesincreasedproteinexpressionofGABAARsincorticalneurons,butGABAAR-mediatedwhole-cellcurrentsarenotpotentiatedbyFMRPtransfection.TheseresultssuggestthepossibilitythatintracellularsignalingantagonizingGABAARactivitymayplayaroleininhibitingGABAARfunctioninFMRP-transfectedneurons.WefurthershowthatFMRPtransfectionresultsinanenhancedproteinexpressionofPTEN,whichcontributestotheinhibitionofGABAARfunctioninFMRP-transfectedneurons.TheseresultsindicatethatGABAARsareregulatedbyFMRPthroughbothanup-regulationofGABAARexpressionandaPTENenhancement-inducedinhibitionofGABAARfunction,suggestingthatanabnormalregulationofGABAARandPTENbythelossofFMRPunderliesthepathogenesisofFXS.(IJPPP1307006).
