The effect of topical capsaicin-induced sensitization on heat-evoked cutaneous vasomotor responses

【 摘 要 】

Brief,localized,cutaneous,non-painfulthermalstimulicanevokeatransientvasomotorresponse,causingincreasedcutaneousbloodflowandelevatedskintemperature.Theaimsofthisstudyweretoinvestigate1)ifcutaneoussensitizationbytopicalapplicationofcapsaicin(TRPV1receptoragonist)canfacilitatethesize,durationandspatialextentofthisvasomotorresponseand2)ifmalesandfemalesresponddifferently.Thermalpulses(43#176;Cfor60seconds)wereappliedonleft/rightvolarforearmsof15age-matchedmalesandfemales.Skintemperatureandcutaneousbloodflowweremeasured1,5,10,15,and30minutesafterheatapplicationbeforeandaftertopicalcapsaicin(1%,30minapplication)withcontralateralarmservingasthecontrol.Recordingsweremadefromtheregionofinterestatdistancesof2,4,6,8,and10cmfromthecapsaicinapplicationsite.Sensitizationsignificantlyenhancedskintemperatureforupto30minandcomparedwithnon-sensitizedskinat10min.Femalesshowedthestrongestresponseaftersensitization,buttheresponselastedlongerandspreadmorewidelyinmales.Thebloodflowresponsesweresignificantlylongeraftercapsaicin(from5to30minutesafterthermalapplication).Thisincreasedbloodflowextendedoutsidethetreatedareaupto10minafterstimulation.Aftersensitization,theareaunderthebloodflowresponsecurvesshowedsignificantlystrongerresponsesinfemales,spreading4cmoutsidethestimulationsite.Cutaneoussensitizingcausedprolongedandspatiallyexpandedvasomotorresponsestostandardizedthermalstimulationwithsexspecificdifferences.(IJPPP1305003).
