Two temporal functions of Glass: Ommatidium patterning and photoreceptor differentiation

Muchprogresshasbeenmadeinelucidatingthemolecularnetworksrequiredforspecifyingretinalcells,includingphotoreceptors,butthedownstreammechanismsthatmaintainidentityandregulatedifferentiationremainpoorlyunderstood.Here,wereportthatthetranscriptionfactorGlasshasadualroleinestablishingafunctionalDrosophilaeye.Utilizingconditionalrescueapproaches,weconfirmthatpersistentdefectsinommatidiumpatterningcombinedwithcelldeathcorrelatewiththeoveralldisruptionofeyemorphologyinglassmutants.Inaddition,werevealthatGlassexhibitsaseparableroleinregulatingphotoreceptordifferentiation.Inparticular,wedemonstratetheapparentlossofglassmutantphotoreceptorsisnotonlyduetocelldeathbutalsoafailureofthesurvivingphotoreceptorstocompletedifferentiation.Moreover,thelatereintroductionofGlassinthesedevelopmentallystalledphotoreceptorsiscapableofrestoringdifferentiationintheabsenceofcorrectommatidiumpatterning.Mechanistically,transcriptionprofilingatthetimeofdifferentiationrevealsthatGlassisnecessaryfortheexpressionofmanygenesimplicatedindifferentiation,i.e.rhabdomeremorphogenesis,phototransduction,andsynaptogenesis.Specifically,weshowGlassdirectlyregulatestheexpressionofPph13,whichencodesatranscriptionfactornecessaryforopsinexpressionandrhabdomeremorphogenesis.Finally,wedemonstratetheabilityofGlasstochoreographphotoreceptordifferentiationisconservedbetweenDrosophilaandTribolium,twoholometabolousinsects.Altogether,ourworkidentifiesafundamentalregulatorymechanismtogeneratethefullcomplementofcellsrequiredforafunctionalrhabdomericvisualsystemandprovidesacriticalframeworktoinvestigatethebasisofdifferentiationandmaintenanceofphotoreceptoridentity.
