Thezebrafishfinisanexcellentsystemtostudythemechanismsofdermalbonepatterning.Finraysaresegmentedstructuresthatformsuccessivebifurcationsbothduringontogenesisandregeneration.Previousstudiesshowedthatsonichedgehog(shha)mayregulateregenerativebonepatterningbasedonitsexpressionpatternandfunctionalanalysis.Thepresentstudyinvestigatestheroleoftheshha-expressingcellsinthepatterningoffinraybranches.Theshhaexpressiondomaininthebasalepidermisofeachfinraysplitsintotwopriortoraybifurcation.Inaddition,theosteoblastproliferationprofilefollowsthedynamicexpressionpatternofshha.Azebrafishtransgenicline,2.4shh:gfpABC#15,inwhichGFPexpressionrecapitulatestheendogenousexpressionofshha,wasusedtospecificallyablateshha-expressingcellswithalaserbeam.Suchablationsleadtoadelayinthesequenceofeventsleadingtoraybifurcationwithoutaffectingtheoverallgrowthofthefinray.Theseresultssuggestthatshha-expressingcellsdirectlocalizedosteoblastproliferationandthusregulatebranchingmorphogenesis.Thisstudyrevealsthefinrayasanewaccessiblesystemtoinvestigateepithelialndash;mesenchymalinteractionsleadingtoorganbranching.
