Currentmodelsofleftndash;rightasymmetryholdthatanearlyasymmetricsignalisgeneratedatthenodeandtransducedtolateralplatemesoderminalinearsignaltransductioncascadethroughthefunctionoftheNodalsignalingmolecule.ThePitx2homeoboxgenefunctionsatthefinalstagesofthiscascadetodirectasymmetricmorphogenesisofselectedorgansincludingtheheart.WepreviouslyshowedthatPitx2regulatedanasymmetricpathwaythatwasindependentofcardiacloopingsuggestingasecondasymmetriccardiacpathway.IthasbeenproposedthatinthecardiacoutflowtractPitx2functionsinbothcardiacneuralcrest,asatargetofcanonicalWnt-signaling,andinthemesoderm-derivedcardiacsecondlineage.Weusedfatemapping,conditionallossoffunction,andchimeraanalysisinmicetoinvestigatetheroleofPitx2inoutflowtractmorphogenesis.OurfindingsrevealthatPitx2isdispensableinthecardiacneuralcrestbutfunctionsinsecondlineagemyocardiumrevealingthatthiscardiacprogenitorfieldispatternedasymmetrically.
