【 摘 要 】
Mandibulardevelopmentisregulatedbyaninterplaybetweenaspecifiedbranchialarchectodermandaplasticmesenchyme.Moreover,signalingfromthepharyngealendodermhasbeenshowntobeimportantformandibularmorphogenesis.Togaininsightintothemechanismsregulatingmandibularpattern,itisimportanttoinvestigatethefunctionoftheepithelial-derivedsignals.Bmp4isexpressedinbothdistal,mandibulararchectodermandpharyngealendoderm.Here,weshowthatdeletionofBmp4inthemandibularectodermandtoalesserextentinthepharyngealendoderm,resultedinseveredefectsinmandibulardevelopment.Furthermore,ourdatauncovereddifferentBmp4thresholdsforexpressionoftheBmp-dependentMsx1andMsx2genesinmandibularmesenchyme.WealsofoundthatectodermalFgf8expressionwasbothactivatedandrepressedbyBmp4inadosage-dependentfashionindicatinganovelBmp4functioninthreshold-specificregulationofFgf8transcription.Lastly,weprovideevidencethatPrxhomeoboxgenesrepressexpressionofanMsx2transgene,previouslyshowntobeBmp4-responsive,revealingamechanismfordifferentialregulationofMsx1andMsx2byBmpsignaling.
