Inthedevelopinglimb,Bmp4isexpressedintheapicalectodermalridge(AER)andunderlyingmesoderm.InsightintothefunctionofBmp4inlimbdevelopmenthasbeenhamperedbytheearlyembryoniclethalityofBmp4nullembryos.WedirectlyinvestigatedBmp4usingaconditionalnullalleleofBmp4andthePrx1cretransgenetoinactivateBmp4inlimbbudmesoderm.ThelimbbudmesodermofPrx1cre;Bmp4mutantswasdefectiveinproductionofBmp4butstillcompetenttorespondtoBmpsignaling.Prx1cre;Bmp4mutantembryoshaddefectivedigitpatterningincludinghindlimbpreaxialpolydactylywithposteriordigittransformations.ThePrx1cre;Bmp4mutantsalsohadpostaxialpolydactylywithdigitfiveduplications.Bmp4mutantlimbshaddelayedinductionandmaturationoftheAERthatresultedinexpandedShhsignaling.Moreover,theAERpersistedlongerintheBmp4mutantlimbbudsexposingtheformingdigitstoprolongedFgf8signaling.OurdatashowthatBmp4inlimbmesodermregulatesAERinductionandmaturationandimplicatesignalingfromtheAERinregulationofdigitnumberandidentity.
