Inhibition of myoblast migration via decorin expression is critical for normal skeletal muscle differentiation

Duringlimbskeletalmuscleformation,committedmusclecellsproliferateanddifferentiateinthepresenceofextracellularsignalsthatstimulateorrepresseachprocess.Proteoglycansareextracellularmatrixorganizersandmodulatorsofgrowthfactoractivities,regulatingmuscledifferentiationinvitro.Previously,wecharacterizedproteoglycanexpressionduringearlylimbmuscleformationandshowedaspatiotemporalrelationbetweentheonsetofmyogenesisandtheexpressionofdecorin,animportantmuscleextracellularmatrixcomponentandpotentregulatorofTGF-βactivity.Toevaluatedecorinrsquo;sroleduringinvivodifferentiationincommittedmusclecells,wegraftedwildtypeanddecorin-nullmyoblastsontochicklimbbuds.Theabsenceofdecorinenhancedthemigrationanddistributionofmyoblastsinthelimb,correlatingwiththeinhibitionofskeletalmuscledifferentiation.Bothphenotypeswererevertedbydenovodecorinexpression.Invitro,wedeterminedthatbothdecorincoreproteinanditsglycosaminoglycanchainwererequiredtoreversethemigrationphenotype.Resultspresentedheresuggestthattheenhancedmigrationobservedindecorin-nullmyoblastsmaynotbedependentonchemotacticgrowthfactorsignalingnorthedifferentiationstatusofthecells.Decorinmaybeinvolvedintheestablishmentand/orcoordinationofacriticalmyoblastdensity,throughinhibitionofmigration,thatpermitsnormalmuscledifferentiationduringembryonicmyogenesis.
