Themechanismsthatcontrolproliferationanddifferentiationofembryoniclungmesenchymearelargelyunknown.WedescribeanexplantsysteminwhichexogenousrecombinantN-SonicHedgehog(N-Shh)proteinsustainsthesurvivalandproliferationoflungmesenchymeinadose-dependentmanner.Inaddition,Shhupregulatesseveralmesenchymalcellmarkers,includingitstargetgenePatched(Ptc),intercellularsignalinggenesBoneMorphogeneticProtein-4(Bmp4)andNoggin(Nog),andsmoothmuscleactinandmyosin.InexplantsexposedtoN-Shhinthemedium,theseproductsareupregulatedthroughoutthemesenchyme,butnotintheperiphery.Thisexclusionzonecorrelateswiththepresenceofanoverlyingmesotheliallayer,which,asinvivo,expressesFibroblastGrowthFactor9(Fgf9).RecombinantFgf9proteininhibitsthedifferentiationresponseofthemesenchymetoN-Shh,butdoesnotaffectproliferation.Weproposeamodelforhowfactorsmadebytwoepithelialcellpopulations,theinnerendodermandtheouterjacketofmesothelium,coordinatelyregulatetheproliferationanddifferentiationofthelungmesoderm.
