Neuronal Differentiation from Postmitotic Precursors in the Ciliary Ganglion

【 摘 要 】

Inthechickciliaryganglion,neuronalnumberiskeptconstantbetweenSt.29andSt.34(E6–E8)despitealargeamountofcelldeath.Here,wecharacterizethesourceofneurogeniccellsintheganglionasundifferentiatedneuralcrest-derivedcells.AtSt.29,neuronsandnonneuronalcellsintheciliaryganglionexpressedtheneuralcrestmarkersHNK-1andp75NTR.Over50%ofthecellswereneuronsatSt.29;ofthenonneuronalcells,asmallpopulationexpressedglialmarkers,whereasthemajoritywasundifferentiated.Whenplacedinculture,nonneuronalcellsacquiredimmunoreactivityforHuD,suggestingthattheyhadcommencedneuronaldifferentiation.Thenewlydifferentiatedneuronsarosefromprecursorsthatdidnotincorporatebromodeoxyuridine.Totestwhethertheseprecursorscouldundergoneuraldifferentiationinvivo,purifiednonneuronalcellsfromSt.29quailgangliaweretransplantedintochickembryosatSt.9–14.Subsequently,quailcellsexpressingneuronalmarkerswerefoundinthechickciliaryganglion.TheexistenceofthisprecursorpoolwastransientbecausenonneuronalcellsisolatedfromSt.38gangliafailedtoformneurons.SinceallciliaryganglionneuronsarebornpriortoSt.29,theseresultsdemonstratethattherearepostmitoticneuralcrest-derivedprecursorsinthedevelopingciliaryganglionthatcandifferentiateintoneuronsintheappropriateenvironment.
