【 摘 要 】

Maturingamphibianoocytesundergodrasticmorphologicalchanges,includinggerminalvesiclebreakdown(GVBD),chromosomecondensation,andspindleformationinresponsetoprogesterone.Twokinases,maturation-promotingfactor(MPF)andmitogen-activatedproteinkinase(MAPK),areinvolvedinthesechanges,buttheirpreciserolesareunknown.UnlikeinXenopusoocytes,discriminationofthefunctionsofMAPKandMPFinRanaoocytesiseasyowingtothelackofpre-MPF.WeinvestigatedtherolesofthesekinasesbycarefulobservationsofchromosomesandmicrotubulesinRanaoocytes.MPFandMAPKactivitiesweremanipulatedbytreatmentwithprogesterone,c-mosmRNA,orcyclinBmRNAincombinationwithMAPKkinaseinhibitors.Activationofonekinasewithoutactivationoftheotherinducedonlylimitedevents;GVBDwasinducedbyMPFwithoutMAPK,andreorganizationofmicrotubulesatGVBDwasinducedbyMAPKwithoutMPF,butothereventswerenotinduced.Incontrast,coactivationofMPFandMAPKbyinjectionofc-mosandcyclinBmRNApromotedalmostallofthemorphologicalchangesthatoccurduringmaturationwithoutprogesterone,indicatingthatthesearecontrolledbycooperationofMPFandMAPK.TheresultsrevealedthefunctionsofMAPKandMPFineachprocessofsequentialmorphologicalchangesduringoocytematuration.
