【 摘 要 】
Theprocessofendochondralossificationinwhichthebonesofthelimbareformedaftergenerationofcartilagemodelsisdependentonapreciselyregulatedprogramofchondrocytematuration.Here,weshowthatthehomeobox-containinggeneDlx5isexpressedattheonsetofchondrocytematurationduringtheconversionofimmatureproliferatingchondrocytesintopostmitotichypertrophyingchondrocytes,acriticalstepinthematurationprocess.Moreover,retroviralmisexpressionofDlx5duringdifferentiationoftheskeletalelementsofthechicklimbinvivoresultsintheformationofseverelyshortenedskeletalelementsthatcontainexcessivenumbersofhypertrophyingchondrocyteswhichextendintoectopicregions,includingsitesnormallyoccupiedbyimmaturechondrocytes.Theexpansionintheextentofhypertrophicmaturationdetectablehistologicallyisaccompaniedbyexpandedandupregulateddomainsofexpressionofmolecularmarkersofchondrocytematuration,particularlytypeXcollagenandosteopontin,andbyexpansionofmineralizedcartilagematrix,whichischaracteristicofterminalhypertrophicdifferentiation.Furthermore,Dlx5misexpressionmarkedlyreduceschondrocyteproliferationconcomitantwithpromotinghypertrophicmaturation.Takentogether,theseresultsindicatethatDlx5isapositiveregulatorofchondrocytematurationandsuggestthatitregulatestheprocessatleastinpartbypromotingconversionofimmatureproliferatingchondrocytesintohypertrophyingchondrocytes.RetroviralmisexpressionofDlx5alsoenhancesformationofperiostealbone,whichisderivedfromtheDlx5-expressingperichondriumthatsurroundsthediaphysesofthecartilagemodels.ThissuggeststhatDlx5maybeinvolvedinregulatingosteoblastdifferentiation,aswellaschondrocytematuration,duringendochondralossification.
