【 摘 要 】
Trophicfactorsareaheterogeneousgroupofmoleculesthatpromotecellgrowthandsurvival.Infreshwaterplanarians,thesmallsecretedproteinTCEN49islinkedtotheregionalmaintenanceoftheplanariancentralbodyregion.Toinvestigateitsfunctioninvivo,weperformedloss-of-functionandgain-of-functionexperimentsbyRNAinterferenceandbytheimplantationofmicrobeadssoakedinTCEN49,respectively.WeshowthatTCEN49behavesasatrophicfactorinvolvedincentralbodyregionneuronsurvival.Inplanariantailregenerates,tcen49expressioninhibitionbydouble-strandedRNAinterferencecausesextensiveapoptosisinvariouscelltypes,includingnervecells.ThisphenotypeisrescuedbytheimplantationofmicrobeadssoakedinTCEN49afterRNAinterference.Ontheotherhand,inorganismscommittedtoasexualreproduction,bothtcen49mRNAanditsproteinaredetectednotonlyinthecentralbodyregionbutalsointheposteriorregion,expandingfromcellsclosetotheventralnervechords.Insomecases,theimplantationofmicrobeadssoakedinTCEN49intheposteriorbodyregiondrivesorganismstoreproduceasexually,andtheinhibitionoftcen49expressionobstructsthisprocess,suggestingalinkbetweenthecentralnervoussystem,TCEN49,regionalinduction,andasexualreproduction.Finally,thedistributionofTCEN49cysteineandtyrosineresiduesalsopointstoacommonevolutionaryoriginforTCEN49andmolluscanneurotrophins.
