【 摘 要 】
Vasaisawidelyconservedgermlinemarker,bothinvertebratesandinvertebrates.Weidentifyavasaorthologue,Sgvasa,anduseittostudygermlinedevelopmentinthegrasshopperSchistocercagregaria,aspeciesinwhichnogermplasmhasbeenidentified.Inadults,Sgvasaisspecificallyexpressedintheovaryandtestis.Itisexpressedathighlevelsduringearlyoogenesis,butnodetectablevasaRNAandlittleVasaproteinarepresentinmatureunlaideggs.Noneappearstobelocalizedtoanydefinedregionoftheeggcortex,suggestingthatgermlinespecificationmaynotdependonmaternalgermplasmexpressingvasa.Vasaproteinisexpressedinmostcleavageenergidsastheyreachtheeggsurfaceandpersistsathighlevelsinmostcellsaggregatingtoformtheembryonicprimordium.However,aftergastrulation,Vasaproteinpersistsonlyinextraembryonicmembranesandincellsattheoutermarginofthelateheart-stageembryo.Intheembryo,itthenbecomerestrictedtocellsatthedorsalmarginoftheformingabdomen.Inolderembryos,theseVasa-positivecellsmovetowardthemidline;Vasaproteinaccumulatesasymmetricallyintheircytoplasm,apatterncloselyresemblingthatofgermcellsinlateembryonicgonads.Thus,wesuggestthattheVasa-stainedcellsintheabdominalmarginaregermcells,asproposedbyNelson(1934),andnotcardioblasts,ashasbeenproposedbyothers.
