Thecontributionsthatcontrolofcellproliferationandcellgrowthmaketodevelopmentalregulationoforganandbodysizeremainpoorlyexplored,particularlywithrespecttoendocyclesinpolyploidtissues.TheepitheliumofthemarinechordateOikopleuradioicaiscomposedofafixednumberofcellsgroupedinterritoriesaccordingtogene-specificexpressionandnuclearsizesandshapes.Astheanimalgrows10-foldduringthelifecycle,epithelialcellsincreaseinsizedifferentiallyasafunctionoftheirspatialposition.Weshowthatthiscellularpatternreflecteddifferencesinploidylevelsrangingfrom34to1300C.Thediverseploidylevelsindefinedcellularfieldsresultedbothfromdifferenttimingofentryintoendocyclesandfromcell-specificregulationofendocyclelengths.Throughoutthelifecycle,differentialcellsizeandploidyincreaseswereaccompaniedbyfield-specificprofilesofprogressivereductionsinG-phaseduration.Endocycleswereasynchronousamongcellsofagivenepithelialterritory,butattheresolutionofindividualcells,bothDNAreplicationtimingandploidylevelswerebilaterallysymmetric.Thetransparent,accessible,oikoplasticepitheliumisamodelofchoiceforthestudyofendoreduplicationinthecontextofpatternformationandgrowth.
