Duringdevelopment,motorandsensoryaxonsgrowtoperipheraltargetswithremarkableprecision.Whereasmuchhasbeenlearnedaboutthedevelopmentofmotoneuronconnectivity,lessisknownabouttheregulationofcutaneousinnervation.Inadults,dorsalrootganglia(DRG)innervatecharacteristicskinregions,termeddermatomes,andtheiraxonsprojectsomatotopicallyinthedorsalhorn.Here,wehaveinvestigatedwhethercutaneousneuronsareselectivelymatchedwithspecificskinregions,andwhetherperipheraltargetskininfluencesthecentralconnectionsofcutaneousneurons.Toaddressthesequestions,weshiftedlimbbudsrostrallyinchickembryospriortoaxonoutgrowth,causingDRGstoinnervatenovelskinregions,andmappedtheresultingdermatomesandcentralprojections.Followinglimbshifts,cutaneousinnervationarosefrommorerostralandfromfewerDRGsthannormal,buttheoveralldermatomepatternwaspreserved.Thus,DRGsparceloutinnervationofskininaconsistentmanner,withnoindicationofmatchingbetweenskinandDRGs.Similarly,cutaneousnervesestablisheda“normal”somatotopicmapinthedorsalhorn,butinmorerostralsegmentsthanusual.Thus,theperipheraltargetskinmayinfluencethepatternofCNSprojections,butdoesnotdirectcutaneousaxonstospecificpopulationsofneuronsinthedorsalhorn.
