【 摘 要 】

Multipotentneuralstemcells(NSCs)presentinthedevelopingneuraltube(E10.5,neuroepithelialcells;NEP)wereexaminedfortheexpressionofcandidatestemcellmarkers,andtheexpressionofthesemarkerswascomparedwithlaterappearingprecursorcells(E14.5)thatcanbedistinguishedbytheexpressionofembryonicneuralcelladhesionmolecule(E-NCAM)andA2B5.NEPcellspossessgapjunctions,expressconnexins,andappeartolacklongcilia.Mostcandidatemarkers,includingNestin,Presenilin,Notch,andNumb,wereexpressedbybothNEPcellsaswellasothercellpopulations.Fibroblastgrowthfactorreceptor4(FGFR4),Frizzled9(Fz9),andSRYbox-containinggene2(Sox2)asassessedbyimmunocytochemistryandinsituhybridizationaremarkersthatappeartodistinguishNSCsfromotherprecursorcells.NeitherHoechst33342norrhodamine-123staining,telomerase(Tert)expression,telomeraseactivity,orbreakpointclusterregionprotein1(Bcrp1)transporterexpressioncouldbeusedtodistinguishNEPstemcellsfromotherdividingcells.NEPcells,however,lackedexpressionofseverallineagemarkersthatareexpressedbylaterappearingcells.TheseincludedabsenceofexpressionofCD44,E-NCAM,A2B5,epidermalgrowthfactorreceptor(EGFR),andplatelet-derivedgrowthfactorreceptor-alpha(PDGFRα),suggestingthatnegativeselectionusingcellsurfaceepitopescouldbeusedtoisolatestemcellpopulationsfrommixedculturesofcells.UsingmixedculturesofcellsisolatedfromE14.5stageembryos,weshowthatNEPcellscanbeenrichedbydepletingdifferentiatingcellsthatexpressE-NCAMorA2B5immunoreactivity.Overall,ourresultsshowthataspectrumofmarkersusedincombinationcanreliablydistinguishmultipotentNSCsfromotherprecursorcellsaswellasdifferentiatedcellspresentintheCNS.
