Cardiacneuralcrestcellsareessentialfornormaldevelopmentofthegreatvesselsandtheheart,givingrisetoarangeofcelltypes,includingbothneuronalandnon-neuronaladventitialcellsandsmoothmuscle.Endothelin(ET)signalingplaysanimportantroleinthedevelopmentofcardiacneuralcrestcelllineages,yettheunderlyingmechanismsthatacttocontroltheirmigration,differentiation,andproliferationremainlargelyunclear.Weexaminedtheexpressionpatternsofthereceptor,ETA,andtheET-specificconvertingenzyme,ECE-1,inthepharyngealarchesandgreatvesselsofthedevelopingchickembryo.Insituhybridizationanalysisrevealedthat,whileETAisexpressedinthepharyngealarchmesenchyme,populatedbycardiacneuralcrestcells,ECE-1expressionislocalizedtotheoutermostectodermalcellsofthearchesandthentotheinnermostendothelialcellsofthegreatvessels.ThisdynamicpatternofexpressionsuggeststhatonlyasubpopulationofneuralcrestcellsintheseregionsisresponsivetoETsignalingatparticulardevelopmentaltimepoints.Totestthis,retroviralgenedeliverywasusedtoconstitutivelyexpresspreproET-1,aprecursorofmatureET-1ligand,inthecardiacneuralcrest.Thisresultedinaselectiveexpansionoftheoutermost,adventitialcellpopulationinthegreatvessels.Incontrast,neitherdifferentiationnorproliferationofneuralcrest-derivedsmoothmusclecellswassignificantlyaffected.TheseresultssuggestthatconstitutiveexpressionofexogenouspreproET-1inthecardiacneuralcrestresultsinexpansionrestrictedtoanadventitialcellpopulationofthedevelopinggreatvessels.
