Thereceptortyrosinekinase(RTK)signalingnetworkplaysacentralroleinregulatingcellulardifferentiation,proliferation,andsurvivalinallmetazoananimals.ExcessiveorcontinuousactivationoftheRTKpathwayhasbeenlinkedtocarcinogenesisinmammals,underscoringtheimportanceofpreventinguncontrolledsignaling.ThisreviewwillfocusontheinhibitorymechanismsthatkeepRTK-mediatedsignalsincheck,withemphasisonconservedprinciplesdiscernedfromstudiesusingDrosophilaasamodelsystem.Twogeneralstrategiesofinhibitionwillbediscussed.Thefirst,thresholdregulation,postulatesthataneffectivewayofantagonizingRTKsignalingistoerectandmaintainhighthresholdbarriersthatpreventinappropriateresponsestomoderatesignalinglevels.Activationofthepathwayabovethislevelovercomestheinhibitoryblocksandshiftsthebalancetoallowapositiveflowofinductiveinformation.Asecondlayerofnegativeregulationinvolvinginductionofnegativefeedbackloopsthatlimittheextent,strength,ordurationofthesignalpreventsrunawaysignalinginresponsetothehighlevelsofactivationrequiredtosurmountthethresholdbarriers.Suchautoinhibitorymechanismsattenuatesignalingatcriticalpointsthroughoutthenetwork,fromthereceptortothedownstreameffectors.
