【 摘 要 】
Theseacucumber,Holothuriaglaberrima,hasthecapacitytoregenerateitsinternalorgans.Intestinalregenerationisaccomplishedbythethickeningofthemesentericborderandtheinvasionofthisthickeningbymucosalepitheliumfromtheesophagusandthecloaca.Extracellularmatrix(ECM)remodelinghasbeenassociatedwithmorphogeneticeventsduringembryonicdevelopmentandregeneration.WehaveusedimmunohistochemicaltechniquesagainstECMcomponentstoshowthatdifferentialchangesoccurintheECMduringearlyregeneration.Labelingoffibrouscollagenouscomponentsandmuscle-relatedlaminindisappearfromtheregeneratingintestineandmesentery,whilefibronectinlabelingand4G7(anechinodermECMcomponent)arecontinuouslypresent.Westernblotsconfirmadecreaseinfibrouscollagencontentduringthefirst2weeksofregeneration.Wehavealsoidentifiedfive1,10-phenanthroline-sensitivebandsincollagengelatinzymographs.Thegelatinolyticactivitiesofthesebandsareenhancedduringearlystagesofregeneration,suggestingthatthemetalloproteaseactivityisassociatedwithECMremodeling.InhibitionofMMPsinvivowith1,10-phenanthroline,p-aminobenzoyl-Gly-Pro-D-Leu-D-AlahydroxamateorN-CBZ-Pro-Leu-GlyhydroxamateproducesareversibleinhibitionofintestinalregenerationandECMremodeling.OurresultsshowthatsignificantchangesinECMcontentoccurduringintestineregenerationintheseacucumberandthattheonsetofthesechangesiscorrelatedtotheproteolyticactivitiesofMMPs.
