Requirement for Both EDEN and AUUUA Motifs in Translational Arrest of Mos mRNA upon Fertilization of Xenopus Eggs

【 摘 要 】

TranslationalarrestofmaternalMosmRNAuponfertilizationofXenopuseggsisaprerequisitefortheinitiationofembryonicdivisions.Recentstudiessuggestthatanembryodeadenylationelement(EDEN)presentinthe3′untranslatedregion(3′UTR)issufficientfordeadenylation(and,hence,probablyfortranslationalarrest)ofMosmRNAafterfertilization.BydirectlymonitoringtranslationofnumerousMosmRNAconstructsinXenopuseggs,however,weshowherethattheEDENisnecessarybutnotsufficientfortranslationalarrestofMosmRNA.WedemonstratethattwoAUUUAmotifs,eachlocatedsolitarilyanddistantlyfromtheEDEN,arealsorequiredforthetranslationalarrestofMosmRNAafterfertilization.Significantly,translationalarrestofEg2mRNA,anotherEDEN-containingmaternalmRNA,alsorequiresasingleAUUUAmotiflocatedfarfromtheEDEN.Analysisofthepoly(A)tailsofvariousMosmRNAconstructsindicatesthattheEDENaloneconfersonlypartialdeadenylationonMosmRNA,andthattheAUUUAmotifsacttoenhanceEDEN-directeddeadenylationinaposition-dependentmanner.Finally,introductionofanexcessoftheEDEN,butnottheAUUUAmotifs,intoeggscanrestoretranslationofendogenousMosmRNA.TheseresultssuggestthattheEDEN,onlytogetherwithappropriatelypositionedAUUUAmotifsandatrans-actingfactor(s),canefficientlydeadenylateandhencetranslationallyarrestMos(aswellasEg2)mRNAafterfertilization.
