【 摘 要 】
Ductalbuddinginthedevelopingprostateisatestosterone-dependenteventthatinvolvessignalingbetweentheurogenitalsinusepithelium(UGE)andurogenitalsinusmesenchyme(UGM).WeshowherethatductalbudformationisassociatedwithfocusedexpressionofSonichedgehog(Shh)intheepitheliumofnascentprostatebudsandinthegrowingtipsofelongatingprostateducts.ThispatternoflocalizedShhexpressionoccursinresponsetotestosteronestimulation.ThegenefortheShhreceptor,Ptc1,isexpressedintheUGM,asarethemembersoftheGligenefamilyoftranscriptionalregulators(Gli1,Gli2,andGli3).ExpressionofPtc1,Gli1,andGli2islocalizedprimarilytomesenchymesurroundingprostatebuds,whereasGli3isexpresseddiffuselythroughouttheUGM.AstrongdependenceofGli1(andPtc1)expressiononShhsignalingisdemonstratedbyinductionofexpressioninboththeintacturogenitalsinusandtheisolatedUGMbyexogenousSHHpeptide.AsimilardependenceofGli2andGli3expressiononShhisnotobserved.Nonetheless,thechemicalinhibitorofShhsignaling,cyclopamine,producedagradedinhibitionofGligeneexpression(Gli1>Gli2>Gli3)inurogenitalsinusexplantsthatwasparalleledbyasevereinhibitionofductalbudding.
