Peripheralnerveandvascularpatternsarecongruentintheadultvertebrate,butthishasbeendisputedinvertebrateembryos.Themostdetailedofthesestudieshaveusedtheavianforelimbasamodelsystem,yetneurovascularanatomicalrelationshipsandcriticalvascularremodelingeventsremaininadequatelycharacterizedinthismodel.Toaddressthis,wehaveusedacombinationofintravascularmarkerinjection,multilabelfluorescentstereomicroscopy,andconfocalmicroscopytoanalyzethespatiotemporalrelationshipsbetweenperipheralnervesandbloodvesselsintheforelimbof818quailembryosfromE2(HH13)toE15(HH41).Wefindthattheneurovascularanatomicalrelationshipsestablishedduringdevelopmentarehighlystereotypicandcongruent.Bloodvesselstypicallyarisebeforetheircorrespondingnerves,butthereareseveralcriticalexceptionstothisrule.Thevascularpatternisextensivelyremodeledfromtheearlieststageexamined(E2;HH13),whereastheperipheralnerves,thefirstofwhichentertheforelimbatE3.5–E4(HH21–HH24),haveaprogressivelyunfoldingpatternthat,onceformed,remainsessentiallyunchanged.TheadultneurovascularpatternisnotestablisheduntilE8(HH34).Peripheralnervesarealwaysfoundtotrackcloseandparalleltothevasculature.Astheytrackdistally,peripheralnervesalwayslieonthesideofthevasculatureawayfromthecenteroftheforelimb.Neurovascularpatternshaveahierarchyofcongruencethatishighestinthedorsoventralplane,followedbytheanteroposterior,andlastlytheproximodistalplanes.
