【 摘 要 】

Membersofthebasichelix–loop–helix(bHLH)transcriptionfactorfamilyplayanessentialroleinmultipledevelopmentalprocesses.Duringneurogenesis,positiveandnegativeregulationbybHLHproteinsisessentialforproperdevelopment.HerewereporttheidentificationandinitialcharacterizationofthebHLHgene,Neuronaltwist(N-twist),namedforitsneuralexpressionpatternandhighsequencehomologyandphysicallinkagetothemesodermalinhibitor,M-twist.N-twistisexpressedinthedevelopingmousecentralnervoussysteminthemidbrain,hindbrain,andneuraltube.Thisneuralexpressionisconservedininvertebrates,asexpressionoftheDrosophilaorthologofN-twistisalsorestrictedtothecentralnervoussystem.LikeotherbHLHfamilymembers,N-TwistheterodimerizeswithEproteinandbindsDNAataconsensusbHLH-bindingsite,theEbox.WeshowthatN-TwistinhibitsMASH1-dependenttranscriptionalactivationbysequesteringEproteininadominantnegativefashion.Thus,thesestudiessupportthenotionthatN-Twistrepresentsanovelnegativeregulatorofneurogenesis.
