【 摘 要 】
Oligodendrocytesareproducedfromthesameregionoftheventralspinalcordthatearliergeneratedmotorneuronsinbirdandrodentembryos.MotorneuronandoligodendrocyteprecursorcellsexpressOliggenes,whichencodebasichelix–loop–helixtranscriptionfactorsthatplayimportantrolesinthedevelopmentofbothmotorneuronsandoligodendrocytes.Wefoundthatoligodendrocytesdevelopsimilarlyinzebrafishembryos,inthattheyarisefromventralspinalcordandmigratetonewpositions.Developingprimarymotorneuronsandoligodendrocytesexpressolig2asdoneuralplatecellsthatgiverisetobothprimarymotorneuronsandoligodendrocytes.Lossofolig2functionpreventedprimarymotorneuronandoligodendrocytedevelopment,whereasolig2overexpressionpromotedformationofexcessprimarymotorneuronsandoligodendrocytes.WeprovidegeneticevidencethatHedgehogsignalingisrequiredforzebrafisholig2expressionandoligodendrocytedevelopment.However,olig2overexpressiondidnotpromoteprimarymotorneuronoroligodendrocytedevelopmentinembryoswithreducedHedgehogsignalingactivity.OnepossibilityconsistentwiththesedataisthatHedgehogsignaling,partlybyinducingolig2expression,specifiesneuralprecursorcellsthathavepotentialforprimarymotorneuronoroligodendrocytefate.
