【 摘 要 】

Duringvertebrateeyedevelopment,theopticvesicleoriginatingfromtheneuroectodermispartitionedintoadomainthatwillgiverisetotheneuralretina(NR)andanotherthatwillgiverisetotheretinalpigmentedepithelium(RPE).PreviousstudieshaveshownthatectopicexpressionofFGFsintheRPEinducesRPE-to-NRtransdifferentiation.Similarly,anaturallyoccurringmutationofthetranscriptionfactorMitfinmouseresultedintheformationofasecondneuralretinainplaceofthedorsalRPE,buttheputativesignalingpathwaylinkingFGFtoMitfregulationispresentlyunknown.Inculturesofneuralcrest-derivedmelanocytes,theMAPKpathwaywasrecentlyshowntotargettheMitftranscriptionfactorforubiquitin-dependentproteolysis,resultinginarapiddegradationanddownregulation.Inthepresentstudy,weshowthatectopicexpressionofaconstitutivelyactivatedalleleofMEK-1,theimmediateupstreamactivatoroftheMAPKERK,inchickenembryonicretinainovo,inducestransdifferentiationoftheRPEintoaneural-likeepitheliumthatiscorrelatedwithadownregulationofMitfexpressioninthepresumptiveRPE.
