Kidneyepitheliadevelopfromthemetanephricmesenchymeafterreceivinginductivesignalsfromtheuretericbudandfromtherenalstroma.However,itisnotclearhowthesesignalsinducethedifferenttypesofepitheliathatmakeupthenephron.Toinvestigateinductivesignaling,wehaveisolatedclustersofepithelialprogenitorsfromthemetanephricmesenchyme,therebyseparatingthemfromtherenalstroma.WhentheisolatedprogenitorsweretreatedwiththeuretericbudfactorLIF,theyexpressedepithelialproteins(ZO-1,E-cadherin,lamininα5)andproducednephrons(36glomeruliwith58tubules),indicatingthattheyarethetargetofinductivesignalingfromtheuretericbud,andthatrenalstromaisnotabsolutelyrequiredforepithelialdevelopmentinvitro.Infact,stroma-depletedepithelialprogenitorsproducedsevenfoldmoreglomerulithandidintactmetanephricmesenchyme(5glomeruli,127tubules).Conversely,whenepithelialprogenitorsweretreatedwithbothLIFandproteinssecretedfromarenalstromalcellline,glomerulogenesiswasabolishedbuttubularepitheliawereexpanded(0glomeruli,47tubules).Hence,byisolatingepithelialprogenitorsfromthemetanephricmesenchyme,weshowthattheyaretargetedbyfactorsfromtheuretericbudandfromtherenalstroma,andthatepithelialdiversificationisstimulatedbytheuretericbudandlimitedbyrenalstroma.
