Mostregulatorygenesareemployedmultipletimestocontroldifferentprocessesduringdevelopment.TheDrosophilaOvo/Shavenbaby(Svb)transcriptionfactorisrequiredbothforgermlineandepidermaldifferentiation,tworolesalsofoundforitsorthologm-ovo1inmice.InDrosophila,thesetwodistinctfunctionsarecontributedbyseparatecontrolregionsdirectingtheexpressionofOvo/Svbinthegermline(ovo)andsoma(svb),respectively.WereportherethatalternativesplicingrepresentsanadditionalleveloftheregulationofOvo/Svbfunctionalspecificity.CharacterizationoftheovoD1rv23mutationrevealedthattheintragenicinsertionofanovelretrotransposon,romano,inactivatesovowithoutalteringsvb.Weprovideevidencethatthisinsertiondisruptsagermline-specificalternativeexon,exon2b,whichencodesa178-amino-acidinternalextension(2B).Whilebothisoforms,Ovo+2BandOvo−2B,accumulateduringoogenesis,onlyOvo+2Bisabletofulfillgerminalovofunctions.Ovo−2Bisunable,evenwhenoverexpressed,tofullyrescueoogenicdefectsresultingfromtheabsenceofwildtypeovoproduct.Bycontrast,eitherOvo+2BorOvo−2BgermlineproteincansubstituteforSvbintheepidermis.Ourresultsemphasizethespecificfeaturesofsplicinginthegermline,andrevealitsfunctionalimportanceforthecontrolofovo/svb-dependentovarianandepidermaldifferentiation.
