MoststudiestodateindicatethattheformationoftestiscordsiscriticalforproperSertolicelldifferentiation,inhibitionofgermcellmeiosis,andregulationofLeydigcelldifferentiation.However,theconnectionsbetweentheseeventsarepoorlyunderstood.Theobjectiveofthisstudywastodissectthemolecularandcellularrelationshipsbetweentheseeventsintestisformation.Wetookadvantageofthedifferenteffectsoftwohedgehogsignalinginhibitors,cyclopamineandforskolin,ongonadexplantcultures.BothhedgehoginhibitorsphenocopiedthedisruptiveeffectofDhh−/−onformationoftestiscordswithoutinfluencingSertolicelldifferentiation.However,theyexhibiteddifferenteffectsonothercellulareventsduringtestisdevelopment.TreatmentwithcyclopaminedidnotaffectinhibitionofgermcellmeiosisandmesonephriccellmigrationbutcauseddefectsinLeydigcelldifferentiation.Incontrast,forskolintreatmentinducedgermcellmeiosis,inhibitedmesonephriccellmigration,andhadnoeffectonLeydigcelldifferentiation.Bycarefullycontrastingthedifferenteffectsofthesetwohedgehoginhibitors,wedemonstratethat,althoughformationoftestiscordsanddevelopmentofothercelltypesnormallytakeplaceinatightlyregulatedsequence,eachoftheseeventscanoccurindependentoftheothers.
