Here,wedescribeoneofthemajormaternalregulatorygradients,Dorsal,andthresholdoutputsofgeneexpressionthatresultfromthegradeddistributionofthistranscriptionfactor.TheanalysisofalargenumberofauthenticandsynthetictargetgenessuggeststhattheDorsalgradientdirectlyspecifiesatleastfour,andpossiblyasmanyasseven,differentthresholdsofgeneactivityandtissuedifferentiation.Thesethresholdsinitiatethedifferentiationofthethreeprimaryembryonictissues:themesoderm,neurogenicectoderm,anddorsalectoderm.Moreover,primaryreadoutsoftheDorsalgradientcreateasymmetriesthatsubdivideeachtissueintomultiplecelltypesduringgastrulation.Dorsalpatterningthresholdsrepresenttheculminationofoneofthemostcompletegeneregulationnetworkknownindevelopment,whichbeginswiththeasymmetricpositioningoftheoocytenucleuswithintheeggchamberandleadstothelocalizedactivationoftheToll-Dorsalsignalingpathwayinventralregionsoftheearlyembryo.
