【 摘 要 】

Genesencodingcomponentsofthehedgehogsignalingpathwayaredynamicallyexpressedinthemouseuteruspreparingforimplantation.Indianhedgehog(Ihh),patched(Ptc),andGli3areexpressedatlowlevelsintheendometrialepitheliumonday1ofpregnancy.TranscriptionofIhhincreasesdramaticallyintheluminalepitheliumandglandsfromday3,reachingveryhighlevelsonday4.Overthesameperiod,Ptc,Gli1,Gli2,andnogginarestronglyupregulatedintheunderlyingmesenchymalstroma.TranscriptionofIhhinovariectomizedmiceisinducedbyprogesteronebutnotbyestrogen.LowerinductionofIhh,Ptc,andHoxa10isseeninresponsetoprogesteroneintheuteriofPgr−/−mutantmicelackingprogesteronenuclearsteroidreceptor.ThisfindingsuggeststhatthehormonemayregulateIhhthroughbothnuclearreceptor-dependentand-independentpathways.Wedescribeamethodforculturinguterineexplantsintheabsenceofepithelium.Undertheseconditions,recombinantN-SHHproteinpromotestheproliferationofmesenchymecellsandtheexpressionofnoggin.WeproposethatIHHmadebytheepitheliumnormallyfunctionsasaparacrinegrowthfactorforstromalcellsduringtheearlystagesofpregnancy.
