AcentrallyimportantfactorininitiatingeggactivationatfertilizationisariseinfreeCa2+intheeggcytosol.Inechinoderm,ascidian,andvertebrateeggs,theCa2+riseoccursasaresultofinositoltrisphosphate-mediatedreleaseofCa2+fromtheendoplasmicreticulum.ThereleaseofCa2+atfertilizationinechinodermandascidianeggsrequiresSH2domain-mediatedactivationofaSrcfamilykinase(SFK)andphospholipaseC(PLC)γ.ThoughsomeevidenceindicatesthataSFKandPLCmayalsofunctionatfertilizationinvertebrateeggs,SH2domain-mediatedactivationofPLCγappearsnottoberequired.Muchworkhasfocusedonidentifyingfactorsfromspermthatinitiateeggactivationatfertilization,eitherasaresultofsperm–eggcontactorsperm–eggfusion.Currentevidencefromstudiesofascidianandmammalianfertilizationfavorsafusion-mediatedmechanism;thisissupportedbyexperimentsindicatingthatinjectionofspermextractsintoeggscausesCa2+releasebythesamepathwayasfertilization.
