【 摘 要 】

Mammaryepitheliumcanfunctionallyregenerateupontransplantation.Thisrenewalcapacityhasbeenclassicallyascribedtothefunctionofamultipotentmammaryglandstemcellpopulation,whichhasbeenhypothesizedtobeaprimarytargetintheetiologyofbreastcancer.Severalcomplementaryapproacheswereemployedinthisstudytoidentifyandenrichmammaryepithelialcellsthatretainstemcellcharacteristics.Usinglong-termBrdUlabeling,apopulationoflabelretainingcells(LRCs)thatlackexpressionofdifferentiationmarkershasbeenidentified.LRCsisolatedfrommammaryprimarycultureswereenrichedforstemcellantigen-1(Sca-1)andHoechstdye-effluxing“sidepopulation”properties.Sca-1poscellsinthemammaryglandwerelocalizedtotheluminalepitheliabyusingSca-1+/GFPmice,wereprogesteronereceptor-negative,anddidnotbindpeanutlectin.Finally,theSca-1pospopulationisenrichedforfunctionalstem/progenitorcells,asdemonstratedbyitsincreasedregenerativepotentialcomparedwithSca-1negcellswhentransplantedintotheclearedmammaryfatpadsofhostmice.
